چکیده

Objective: To explore the role of miR-448 in regulating MAGEA6/AMPK signaling pathway biological study hepatocellular carcinoma (HCC) tumor stem cells. Methods: Using database, related expression chips were obtained and regulatory mirnas candidate genes predicted, predicted results analyzed. The effects MAGEA6 on pellet formation rate clone cells detected by immunofluorescence identification cell markers light microscope counting method. migration invasion scratch Transwell assay. Dual luciferase reporter assay to verify whether targets MAGEA6. influence AMPK qRT-PCR Western blot. Results: It was found that may directly regulate Overexpression inhibited characteristics, proliferation, migration, vitro, as well ability xenograft vivo. However, inhibition showed opposite results. In addition, regulates signaling. Silencing adding activator further verified activated targeting MAGEA6, thus affecting hepatoma Conclusions: Our reveal activates thereby is suggested overexpression be a new therapeutic strategy for carcinoma.

Nanocurcumin-Mediated Down-Regulation of Telomerase Via Stimulating TGFβ1 Signaling Pathway in Hepatocellular Carcinoma Cells

Background: Curcumin, extracted from turmeric, represents enormous potential to serve as an anticancer agent. Telomerase is viewed as a prominent molecular target of curcumin, and transforming growth factor-β1 (TGFβ1) has proven to be a major inhibitory signaling pathway for telomerase activity. In the current study, we aimed to explore suppressive effects of nanocurcumin on telomeras...

miR-320 regulates inflammation in EAE through interference with TGF-β signaling pathway

Background: MicroRNAs are small noncoding RNAs that regulate gene expression and involve in many cellular and physiological mechanisems. Recent studies have revealed that dysregulation of microRNAs might contribute to autoimmune disorders such as multiple sclerosis. Based on these findings, we examined the potential role of miR-320 isoforms, miR-320-3p and miR-320-5p, in the context of autoimmu...

MiR-200b/200c/429 subfamily negatively regulates Rho/ROCK signaling pathway to suppress hepatocellular carcinoma metastasis

MiR-200 family is an important regulator of epithelial-mesenchymal transition and has been implicated in human carcinogenesis. However, their expression and functions in human cancers remain controversial. In the work presented here, we showed that miR-200 family members were frequently down-regulated in hepatocellular carcinoma (HCC). Although all five members of miR-200 family inhibited ZEB1/...

Angiopoietin-like protein 8 (betatrophin) may inhibit hepatocellular carcinoma through suppressing of the Wnt signaling pathway

Objective(s): Hepatocellular carcinoma (HCC) is one of the leading fatal neoplasms and the most common primary liver malignancy worldwide. Peptide hormone ANGPTL8 (betatrophin) may act as an important regulator in HCC development through the Wnt/β-catenin pathway. We aimed to evaluate the effects of recombinant ANGPTL8 on Wnt/β-catenin signaling in human liver carcinom...

miR-500a-3p promotes cancer stem cells properties via STAT3 pathway in human hepatocellular carcinoma

BACKGROUND miR-500a-3p has been demonstrated to be involved in the development, progression and metastasis in several human cancers. Constitutive activation of JAK/STAT3 signaling pathway has been reported to play an important role in the development and progression of hepatocellular carcinoma (HCC).The purpose of this study was to determine the biological roles and clinical significance of miR...
